(stat′ĭn) [-stat + -in] Any of the drugs of the class known as 3-hydroxy-3-methylglutaryl coenzyme A (HMG CoA) reductase inhibitors. These drugs have powerful lipid-lowering properties. The names of drugs in this class all end in -statin, e.g., atorvastatin, pravastatin, rosuvastatin, and simvastatin. Drugs from this class reduce the risk of myocardial infarction and stroke. Many statins are metabolized by the cytochrome P450 3A4 isoenzyme of the liver. Drugs that block CYP3A4 (an isoenzyme form of the cytochrome P450 system) enzyme activity, such as erythromycin, clarithromycin, and some agents used to treat HIV infection or hepatitis, may cause muscle and kidney damage when taken with statins.
